摘要
本研究采用饲菌复合失血性休克造成家兔MSOF的实验模型,利用放射配基结合分析法观察了脏器衰竭后24及96小时肝肺组织中β肾上腺素能受体数量和亲和力的改变,并用生化方法监测了同时间点血液及组织中磷酯酶A_2活性的变化。结果表明:家兔出现MSOF后24小时肝组织β受体数量和亲和力变化不大,肺组织β受体数量下降,亲和力不变。MSOF后96小时肝肺β受体数量和亲和力均明显增加。家兔出现MSOF后24及96小时血液及组织中磷酯酶A_2活性逐渐增强,提示β受体的变化及磷酯酶A_2激活在MSOF早晚期病理损伤中起一定作用。
An animal model of multiple system organ failure (MSOF) was develo-ped by hemorrhagic shock plus E coli feeding to rabbits. The Bmax and Kd of Beta-adreneric receptor as Well as phospholipase A_2 activity (PLA_2) in liver and lung tissue ofrabbits with MSOF were measured by radio-ligand binding assay and biochemical method.It was found that the number and affinity of receptor were elevated significantly in liverand lung suffering MSOF for 96 hours, while the activity of PLA_2 was markedly activa-ted in plasma, liver and lung tissue at 24, 96 hours of MSOF. These results suggestedthat the alteration of Beta receptor function and activation of PLA_2 may play a veryimportant role in the late stage of MSOF.
