Na^+-H^+交换阻滞剂对离体鼠心缺血后再灌注损伤的保护作用 被引量:5

摘要 为评价Na^+-H^+交换阻滞剂对缺血再灌注损伤的保护作用,在改良的Langendoff装置上采取主动脉逆灌方法,用成年Wistar大鼠的离体鼠心完全停灌60min,然后100%再灌注30min造成再灌注损伤模型。阿米洛利(0.2、20、100μmol/L)和呋喃苯胺酸(10μmol/L)为药物保护组及不加药物的对照组。结果显示:阿米洛利和呋喃苯胺酸对再灌注时心肌收缩力、冠脉流量的恢复及抑制再灌注心律失常的发生有明显的作用,阿米洛利抗心律失常的效果呈明显的剂量依赖性。因此提示具Na^+-H^+交换阻断作用的阿米洛利和呋喃苯胺酸具明显的抗再灌注损伤的作用。 In order to evaluate the protective effects of Na^+/H^+ exchange blocking agents on ischemic reperfusion injury of the hearts, a model was set up by using a modified Langendoff apparatus and given the heart 60 minutes zero flow perfusion followed by 30 minutes 100%reperfusion. The drug groups were treated with amiloride(0.2, 20, 100 μmol/L) and furosemide (10 μmol/L) and the control group was not treated with drugs. The re- sults showed that during reperfusion: (1)the cardiac contractile force were more than 60% in three amiloride groups(P<0.01)and more than 30% in furosemide group (P<0.05), but was less than 20% in control group; (2) the recovery of coronary flow were more than 60% in three amiloride groups (P<0.01), but in the furosemide group was not much more than control group; (3) mild negative chronotropic effect was observed during later reperfusion of larger amiloride dosage group; (4) the incidence of reperfusion arrhy- thmias were decreased obviously by treatment with amiloride and furosemide; meanwhile amiloride showed dose dependency for anti-reperfused arrhythmias. It is concluded that Na^+/H^+ exchange blocking agents were effective protective agents for post-ischemic reper- fusion injury of the hearts.
