摘要 本文用分光光度法对SD大鼠肺组织脂氧合酶代谢外来化合物的能力及其亚细胞分布进行了研究。在Tris缓冲液pH6.5,13.5mmol亚油酸和210μg胞液蛋白/ml反应溶液的条件下,脂氧合酶对联苯胺等8种化学物有协同氧化作用。去甲二氢愈创木酸等3种酶抑制剂对联苯胺的氧化反应有明显抑制作用。大鼠肺脂氧合酶代谢活性主要位于胞液(占获得总活性的87.6%)。结果提示,脂氧合酶可能是某些外来化合物在肺内代谢的一条重要途径。 The activity of lipoxyenase from SD rat lung to metabo- lize xenobiotics was investigated and the subcellular distribu- tion of the enzyme was examined spectrophotometrically. Un- der the optimum reactive condition (pH6.5 in Tris buffer, 13. 5 m mol linoleic acid and 210μg cytosolic protein/ml reaction media)8 chemicals including benzidine were co-oxidized by the enzyme. Benzidine oxidation was obviously inhibated by NDGA and other 2 lipoxygenase inhibators. The most part of the metabolic activity of rat lung lipoxygenase was found in the cytosolic fraction (87.6% of the total enzyme activity ob- tained). The present data indicated that lipoxygenase may be a important pathway of xenobiotic metabolism in lung.
