弓形虫速殖子抗原组分分析及其免疫反应性的比较研究 被引量:7

摘要 应用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)、酶联免疫印渍试验(ELIB)和酶联免疫吸附试验(ELISA),对四种不同地理株弓形虫-CN、RH、BH、HH速殖子细胞质、细胞膜和代谢抗原组分及株间抗原差异、免疫反应性进行比较研究。ELIB实验结果:RH株高免兔血清与四株细胞质抗原反应,分别出现10、7、4、6条显色的反应区带,其中在35~38kD处都出现极明显的反应区带;与四株细胞膜抗原反应,分别出现5、6、6、9条显色反应区带,其中在31~35kD处都出现极明显的反应区带;与四株代谢抗原反应,分别出现8、7、6、7条反应区带,其中在50kD处、100~108kD处都有明显反应区带。实验结果提示,高免兔血清与四株12种抗原反应结果所出现的区带存在不同程度差异。ELISA检测免疫比值(P/N)大小比较结果表明,无论是细胞质、细胞膜或代谢抗原,在检测人弓形虫抗体阳性血清时,HH、BH较CN、RH的免疫比值高,株间抗原免疫反应似存在一定差异。 By using sodium dodecyl sulfute - polyacrylamidegel electrophoresis (SDS- PAGE), enzyme linked immunoblot (ELIB)and enzyme linked immunosorbent assay (ELISA) , we investigated the components of cytoplasmic, membrane and metabolic antigens of tachyzoites of 4 various geographic strains of Toxoplasma gondii -CN,RH,BH,HH and compared the differences of antigens among the strains. By ELIB;after RH-immuned rabbit sera reacted with 4 cytoplas- mic antigens,there were 10,7,4,6 bands respectively, all the bands appeared in molecular weight regions of 35 and 38 kD. After the sera reacted with 4 membrane antigens, there were 5,6,6,9 bands respectively, the band of 35kD was very obvious. After the sera reacted with 4 metabolic antigens,there were 8,7,6,7 bands respectively, the major fractions were of 50,100 and 108 kD. The result showed that there were remarkable differences among those bands in the reaction of RH-immuned rabbit sera with 12 antigens of 4 strains. The comparision of immune ratio(P/N)in ELISA showed that no matter which antigens were used, cytoplasmic, membrane or metabolic antigens,the P/N of HH and BH in detection of positive sera of human toxoplasmosis antibodies were higher than those of CN and RH. It sugests that there is different antigen immunity among the 4 strains.
