摘要
本文将吲哚花青绿(ICG)用于大鼠移植性肝癌的PDT治疗,通过组织病理学检查及图象分析仪测量肝组织光敏损伤量,发现PDT治疗前给予ICG的实验组光敏坏死量较不给ICG的对照组有明显减少,存活的肿瘤面积无明显差异。说明ICG在PDT治疗中对正常肝细胞有一定的选择性保护作用,而不影响对肿瘤细胞的杀伤作用。观察到ICG对照射表层和靠近中心部的正常肝细胞保护作用不明显;对氩离子激光照射组的保护作用不及对630nm染料激光照射组明显。认为ICG的保护作用受其所在正常肝细胞内含量的限制和光能量分布的影响。
The selective protective effect of indocyanine green (ICG) to hepatocytes in PDTtreated liver with transplantable hepatocarcinoma in rats was studied by histopathological examination, and the mean area, depth, diameter of photosensitized by liver necrosis and remaining survival tumor tissue were measured and analyzed by using an image analyzing system, Twelve rats in each group received HpD and ICG (10 mg/kg, i.v.) 24 h and 10 min prior to laser irradiation, respectively. The tumor in the left lobe was irradiated with 630 nm or 514.5 nm laser beam (380 mW/cm^2, 230 J/cm^2) and the tumor in the central lobe of the same rat was taken as control. The results showed that the values of all parameters of liver necrosis in experimental groups were remarkably decreased than those in coresponding control groups. While the mean area of remaining survived tumor tissue after PDT did not show significant difference between experimental and control groups. It was also found that the protective effect of ICG to the hepatocytes on the superficial layer and the central zone of irradiation was less significant, and the effect of ICG against 514.5 nm laser action was less satisfactory than that of 630 nm laser irradiation. It was considered that the different extent of protective effect of ICG may be due to the different optical properties of 630 and 514.5 nm laser light and the concentration of ICG in the liver tissue.
