摘要
目的了解热疗联合丝裂霉素对大肠癌细胞的影响。方法本文用 MTT 法体外研究热疗联合丝裂霉素(MMC)对人大肠癌细胞株 LOVO 的细胞毒作用。结果单纯热,43℃ 30分钟具有较强的细胞毒作用。单纯化疗,在 MMC 浓度为0.01mg/ml 时,具有较强的细胞毒作用。热疗联合化疗,细胞毒作用明显增强,大于单纯热疗和单纯化疗之和,二者具有协同作用。43℃ 30分钟以上,细胞毒作用很强。单纯热疗和单纯化疗在低渗条件下,细胞毒作用大于等渗的细胞毒作用(P<0.05)。结论经尽可能短的时间,使温度达43℃,持续时间不少于30分钟,MMC 浓度不低于0.01mg/ml 时,热疗联合丝裂霉素对人大肠癌细胞有很强的细胞毒作用。
Objective To study the effect of hyperthermia and MMC on human large intestine cancer cell.Methods The dynamics of cytotoxic effects of continous hyperthermia combined with MMC on human large intestine cancer cell lines LOVO was studied by MTT.Results The cytotoxic effects of hyperthermia alone 43 30min was stronger.When the concentration of MMC was 0.01mg/ml,the cytotoxic effects of chemotherapy alone was stronger,too.Hyperthermia and MMC could be synergetic,the cytotoxic effects of hyperthermia combined with MMC was higher than hyperthermia or MMC alone.The cytotoic effcets of hy- perthermia or MMC alone was higher in hyperthermia than in isosmotic (P<0.05).Conclusion Our data suggested that the cytotoxic effects on human large intestin cancer cells was very large above 0.01mg/ml of concentration of MMC,in isosmotic solution when the temperature rose fast to desired.
