补肾、健脾和活血化瘀方药对去卵巢大鼠骨质疏松的比较性研究 被引量:49

摘要 采用去卵巢大鼠模型,在确定单一因素的条件下比较性研究三种治则对骨代谢的影响。40只4月龄SD♀大鼠,随机分为5组,每组8只。Ⅰ组:假手术组(sham),Ⅱ组:切除卵巢组(OVX),Ⅲ组:切除卵巢加补肾方剂组(OVX+TKR),Ⅳ组:切除卵巢加健脾方剂组(OVX+ISR),Ⅴ组:切除卵巢加活血化瘀中药组(OVX+PBCR)。术后第7天起,Ⅲ~Ⅴ组灌胃中药,Ⅰ~Ⅱ组灌胃蒸馏水。术后97天处死动物。行血清、尿生化检测、胫骨骨组织形态计量学检测和股骨骨密度检测。结果:OVX与sham组相比,胫骨TBV%明显下降(P<0.01),股骨的股骨头、粗隆间、踝间窝上骨密度明显下降(P<0.05~0.01)。OVX+TKR组与OVX组相比,胫骨TBV%明显上升(P<0.01),股骨各部位骨密度均明显升高(P<0.05~0.01),健脾方剂组基本无延缓去势引起的骨量丢失作用。活血化瘀中药有延缓去势引起的大鼠骨量丢失作用。结果提示:三方相比,补肾方剂有显著性对抗去势引起的大鼠骨量丢失作用。 Using ovariectomized rat (OVX),on the condition of single definite factor,the effects of tonifying kidney recipy (TKR),invigorating spleen recipe (ISR) and promoting blood circulation recipy (PBCR) on bone were studied.Fourty female SD rats of four month weighed 230±25g were divided into 5 groups,eight rats a group,group Ⅰ:false operation (sham),group Ⅱ:ovary resection (OVX),group Ⅲ:ovary resection plus TKR(OVX+TKR),group Ⅳ:ovary resection plus ISR(OVX+ISR),group Ⅴ:ovary resection plus PBCR (OVX+PBCR).Seven days after operation,group Ⅲ was given TKR1125g/Kg/d;group Ⅳ was given ISR 3.23g/Kg/d;group Ⅴ was given 2.16g/Kg/d;group Ⅰ and Ⅱ were given only water,five times a week,successively in three months.Ninety seven days after operation,all animals were killed,then following tests were made:biochemical examine of serum and urine,morphologic metrology of tibia,and BMD of femur.Results:compared with group Ⅰ,TBV% of tibia remarkably lowered (P<001),TFS% and TRS% remarkably increased (P<005-001),BMD in neck of femur,intertrochanter,above the intercomdylar fossa remarkably lowered (P<005-001) in group Ⅱ.By all such,OVX rat successfully established a model of osteoporosis,and this model was a highly transform one.Compared with group Ⅱ,in group Ⅲ TBV% of tibia remarkably rose (P<001),TFS% and TRS% remarkably lowered (P<001),BMD in head of femur,neck of femur,intertrochanter,middle segment of femur,above intercondylar fossa of femur remarkably rose (P<005-001),such demonstrated that TKR could remarkably postpone the loses of rat bone mass caused by ovary resection.Compared with group Ⅱ,in group Ⅳ TBV% and TRS% of tibia did not have obvious changes (P>005),TRS% obviously decreased (P<001),BMD in every part of femur did not have distinct discrepancy (P>005),showed that ISR did not have the effect of resisting the loses of bone quantity in rat caused by ovary resection.Compared with group Ⅱ,in group Ⅴ TBV% of the tibia remarkably rose (P<005),TFS% and TFS% remarkably lowered (P<005-001),BMD of the neck of femur remarkably rose (P<005),showed that PBCR had the effects of resisting the loses of bone quantity in rat caused by ovary resection,but the effects were lower than that of the TKR.The results demonstrated that TKR could resist the lose of bone quantity in rat caused by ovary resection,PBCR,also had such effect,but its was lower than that of TKR.
