肿瘤细胞淋巴细胞混合培养诱导产生胶质瘤特异性CTL的实验研究

摘要 目的 通过自身肿瘤激活诱导产生高活性的胶质瘤特异性细胞毒T淋巴细胞(G-S-ChL)。方法应用淋巴细胞、肿瘤细胞混合培养(MLTC)方法,诱导产生G-S-CTL,采用3H-TdR释放法及免疫荧光法检测G-S-CTL的细胞毒活性及免疫表型。结果G—S-CTL与自身LAK相比,抗肿瘤活性大为提高,CD3+、CD4+、CD8+和CD25+细胞明显增多。结论G-S-CTL抗肿瘤活性及免疫表型类似于胶质瘤浸润的淋巴细胞(GIL),对脑胶质瘤过继免疫治疗具有一定的实用价值。 Objective To induce highly active glioma \ specific cy-totoxic T - lymphocyte (G - S - CTL) lines by stimulation of autologous tu-mor cells. Methods G- S - CTL were established by cocultivation of lym-phocytes and autologous tumor cells. The cytotoxic activity and phenotypes of G- S - CTL were detected by 3H - TdR - release assay and immunofluo-rescence. Results Compared with auto - LAK, those G - S - CTL showed much higher antitumor activity; CD3+ 、CD4+、CD8 + and CD25 + cells in-creased evidently. Conclusions These results suggested that the antitumor activity and immunophenotype of G - S-CTL are similar to those of glioma infiltrating lymphocytes (GIL) and have practical value in adoptive im-munotherapy of patiens with brain gliomas.
