摘要
目的观察用肿瘤坏死因子α(TNFα)诱导的人大肠癌 LoVo 细胞凋亡的超微结构特点。方法用体外细胞培养方法,以 TNFα诱导 LoVo 细胞凋亡,倒置显微镜、透射电镜观察细胞形态。结果凋亡早期,LoVo 细胞外形不规则,胞体大小及胞浆变化不大,微绒毛变细,染色质凝集,开始向核膜下集中;中期,胞体缩小,胞膜完整,染色质呈颗粒、块状凝集在核膜内侧、核孔之间;后期,核裂解,形成凋亡小体,并可被未凋亡的肿瘤细胞吞噬和消化。结论 TNFα能够诱导大肠癌 LoVo 细胞凋亡,经透射电镜观察,其具有特征性的形态学改变。
AIM To observe the ultrastructural features of apoptosis occurring in the colorectal cancer LoVo cells induced by tumor necrosis factor α(TNFα).METHODS LoVo cells were cultured in vitro and treated with TNFα,and underwent apoptosis.The morphological changes of a poptosis were observed by the electron microscopy.RESULTS At early stage,the size of cells and plasma didn't change while the microvilli shrunk, the chromatin compacted and gathered;at middle stage, the cells lessened in size,the chromatin appeared granular mass that margined against the nuclear envelope and between nuclear holes;at last stage, the nuclei broke up,apoptotic bodies(Abs)were formed,and Abs might be p hagocytized and digested by the nonapoptotic Lovo cells.CONCLU- SION TNFα can induce apoptosis of human colorectal carcinoma LoVo cells which appeared apoptotive Ultrastructural features.
