摘要
目的研究外源性hPTH(1-34)对新生大鼠成骨细胞基质钙含量及钙化结节形成的影响。方法新生大鼠成骨细胞原代培养,原子吸收分光光度法测细胞基质钙含量,vonKossa染色鉴定钙化结节。结果10-10~10-7mol·L-1hPTH(1-34)持续作用于细胞,基质钙含量与对照相比显著降低;10-8mol·L-1hPTH(1-34)在以48小时为一循环的前12小时给药循环8次时基质钙含量达最高峰,与对照组相比增加576%,且形成钙化结节最多。结论hPTH(1-34)刺激成骨细胞基质钙含量及钙化结节形成与给药方式及药物浓度有关,低浓度间歇性给与hPTH(1-34)能提高成骨细胞基质钙及促进钙化结节形成。
To investigate the effect of human parathyroid hormone(1 34)[hPTH(1 34)]on the content of matrix calcium and bone nodule formation of osteoblasts. Methods Isolating the osteoblasts from newborn rat calvaria through trypsin digestion,contents of matrix calcium were detected by atomic absorptiometry and bone nodules were observed by von Kossa's staining. Results While given the osteoblasts with 10 -10 mol·L -1 hPTH(1 34) to 10 -7 mol·L -1 hPTH(1 34)continuously,the contents of calcium had no discrepancy compared with the control group(P>0 05).Contents of calcium were increased 576% after the first 12 h of each 48 h incubation cycle for 8 cycles compared with control group under the effect of 10 -8 mol·L -1 hPTH(1 34)(P<0 01).The intermittent effect of PTH from 10 -10 to 10 -8 mol·L -1 on osteoblast matrix calcium was directly relative to its concentration.Bone nodules increased greatly under the effect of 10 -8 mol·L -1 hPTH(1 34) after first 12 hours of each 48h incubation cycle for 8 cycles. Conclusions PTH has diverse effect on contents of matrix calcium and bone nodule formation of osteoblasts, depending on the exposure time and its concentration in vitro .Contents of matrix calcium and bone nodule increase after intermittent exposure to hPTH(1 34).
