摘要
目的观察单独和联合使用全反式维甲酸(ATRA)和1,25-二羟维生素D3对大肠癌细胞株LoVo碱性磷酸酶(ALP)活性的变化的影响。方法用不同剂量的ATRA和1,25-二羟维生素D3单独或联合在体外作用于LoVo细胞2、4和6天,用脱氧胆酸钠溶解细胞,动力学方法测定ALP活性,紫外吸收法测定蛋白质含量。结果单独或联合使用ATAR和1,25-二羟维生素D3均可使LoVo细胞的肠型ALP活性逐渐升高,而以联合用药效果更好(P<0.05)。结论联合使用A-TRA和1,25-二羟维生素D3诱导LoVo细胞比单独用药效果更好。
Objective To observe the change of alkaline phosphatases (ALP) activities in large intestinal cancer cells line (LoVo) induced by all trans retinoic acid (ATRA) and/or 1,25-dihydroxy vitamin D 3。 Methods The large intestinal cancer cell line was treated by different dosages of ATRA and/or 1,25-dihydroxy vitamin D 3 for 2,4 or 6 days. Then, the cells were dissolved in sodium deoxycholate. The ALP activities were measured by dynamics assay and the total protein was detected by ultraviolet absorbency method. Results The intestinal type of ALP was gradually increased by drug treatment and the effects of combinative using two drugs were better. Conclusion The differential effects of combinative using ATRA and 1,25 dihydroxy vitamin D 3 on LoVo cells were better than that of using each drug only. It had clinical significance for the diagnosis and treatment on large intestinal cancer.
