摘要
目的 研究血管内皮细胞生长因子(Vascular Endothelial Growth Factor,VEGF)与肝癌细胞转移的相互关系。方法 运用Bodyen Chamber膜侵袭培养系统培养VEGF诱导的肝癌细胞株HepG2.培养结束后,羊膜进行苏木精染色.计算穿过羊膜的下室肝癌细胞数以及停留在羊膜内的肝癌细胞数。结果 用VEGF 1ng/ml,5ng/ml、10ng/ml培养5小时,下室浸润的肝癌细胞数分别为为20、64、38×10^4/ml,分别高于对照组5×10^4/ml.P<0.05或0.015ngmlVEGF培养5小时羊膜中浸润的肝癌细胞数为45个与对照组5个比较,差异有非常显著性,P<0.01。结论 VEGF可以诱导肝癌细胞转移,最佳浓度为5ng/ml0.最适培养时间为2-5小时.VEGF诱导肝癌细胞转移机制与增强肿瘤细胞的迁移能力密切相关。
Objective To study the relationship between Vascular Endothelial Growth Factor(VEGF) and hepatocellular carcinoma metastasis. Methods To have used the Membrane Invasion Culture System(MICS), We cultured the HepG_2 Line induced by VEGF with Bodyen Chamber. At the end of the cultured time, the amnion was stained by Mayer's Hematoxilin, We counted the numbers of cell which go through the membrane from upper part of the Chamber into lower part, and the the numbers of cell which stay inside the membrane. Result The cells in Lower part are respectively 20、64、38×104/ml after cultured 5 hours bylng/ml、5ng/ml、10ng/ml, which is remarkably higher than that which is 5×10~4/ml in control group, P<0.05, or 0.01. The cells in membrane cultured 5 hous by 5ng/ml VEGF are 45, which is dramatically higher than that which is 5 in contral group, P<0.01. Conclution VEGF can induce hepatocellular carcinoma metastasis, The most suitable concentration is 5ng/ml, The most suitable time is about 2-5 hours. The metastasis mechanism caused by VEGF is relative with enhancing migration ability of hepatocellular carcinoma.
