应用抗BrdU单抗观察胃肠道功能紊乱大鼠粘膜细胞增殖动力学变化

摘要 目的:探索胃肠道功能紊乱时胃肠粘膜细胞的增殖能力。方法:应用抗-BrdU的单克隆抗体和免疫组织化学技术分析胃肠道粘膜细胞的增殖情况。结果:BrdU的免疫标记显示,胃肠道功能紊乱时胃肠粘膜细胞的增殖能力下降(P<0.05),经四君子汤预防和治疗的胃肠道功能紊乱模型大鼠胃肠粘膜细胞的增殖力明显增强,与自然恢复大鼠有显著差异(P<0.05)。结论:胃肠道功能紊乱时胃肠粘膜细胞的增殖能力低下,提示胃肠道功能紊乱时全身适应性调节和营养代谢失调,对揭示胃肠道功能紊乱症的本质具有重要意义。 To explore proliferative activity of gastrointestinal mueosal cells in the experimental functional distubance of gastrointestinal tract model. Methods: Proliferation of gastrointestinal mucosal cells was analyzed with monoclonal antibody specific to BrdU and immunohistochemieal techmique. Results: BrdU-immunohistochemical technique demonstrated that proliferation of gastrointestinal mucosal cells was prohibited in the experimental functional disturbance of gastrointestinal tract model. Proliferative activity of gastrointestinal mucosal cells could be increased and even better than spontaneous recovery group after prevention and therapy with Sijunzi decoction. Conclusions: The study implied that general regulation of adaptability and metaholism was in disorder in the experimental functional distuubanee of gastrointestinal tract model and advanced an objective evidence for exploring the exxence of functional disturbance of gastrointestinal tract.
