摘要
本文报道了一适宜人类急性髓系白血病祖细胞(AML-CFU)良好克隆性生长的无血清培养(SFC)体系。采用正交设计,得出主要血清替代物牛血清白蛋白(BSA)、饱和人转铁蛋白(STf)、胆固醇(Chol)、牛胰岛素(Ins)和牛氯化血红素(Hem)浓度依次为0.6%、2×10^(-6)M、2.8μg/ml、15μg/ml和0.06mM。重组人粒/巨噬细胞集落刺激因子(rhGM-CSF)和重组人白介素3(rhIL-3)在SFC体系中的种植率分别为0.776±0.621%和0.574±0.510%((?)±s%),与有血清培养(SCC)体系中的种植率无显著性差异(n=14,P>0.05)。
The paper reports a SFC system supporting optimal clonal growth of human AML-CPU. Under the orthogonal design condition, the combination of the serum substitutes including 0.6% bovine serum albumin
(BSA), 2×10-6M saturated human transfer-tin (STf), 2.8μg/ml cholesterol (Chol), 15μg/ml bovine insulin (Ins)
and 0.06mM hemin (Hem) was critical inour SFC system. Moreover, in SFC the pkting efficiencies obtained by stimulating with recombmant human granulocyte/macrophage colony-stimulating factor (rhGM-CSF) and
recombinant human interleukin-3 (rhIL-3) were 0.776±0.621% and 0.574±0.510% (x+s%), respectively,
which didn't differ significanly from those in serum-containing culture (SCC) (n = 14, P>0.00).
