脑胶质瘤浸润淋巴细胞的分离、培养、NK细胞活性和T细胞表型研究 被引量:2

摘要 本文应用酶消化和不连续密度梯度离心法,从9例原发性脑胶质瘤手术切除的瘤组织中分离出胶质瘤浸润淋巴细胞(GIL),在含IL-2培养液内培养4周,GIL 平均扩增48.4倍,最高达118倍;新制备GIL 的NK 活性极低,经过1周培养,NK 活性明显升高;新制备GIL 中CD_3^+细胞为71.0士11.9%,其中CD_8^+细胞多于CD_4^+细胞。 In the present study,glioma-infiltrating lymphocytes (GIL) were isolated from 9 of 21 surgi-cal biopsy specimens of patients with brain gliomas using enzyme digestion and discontinuousdensity gradient centrifugation.Results indicated that there were a various GIL in 42.8%gliomas.GIL recovered were 1.7 to 6.1×10~5cells/wet gm of glioma.GIL were expanded 48.4-fold on the average and even up to 118-fold by culture in IL-2 medium for a period of 4weeks.NK activity of GIL were strikingly lower than that of GIL in IL-2 culture for a period of1 week.NK activity of normal lymphocyte mimic separation process of GIL was not differenceto that of same lymphocyte.Analysis of surface phenotypes of GIL showed that CD_3^+ cells were71.0±11.9%,CD_4^+ cells were 34.2±6.9% and CD_8^+ cells were 37.0±8.5%.These results suggested that it is hopeful for adoptive immunotherapy of patients with braingliomas to use autologous GIL expanded in the presence of IL-2.
