摘要
将picryl chloride(PC)所致的迟发型变态反应(PC-DTH)导入肝脏诱发肝损伤,方法是将PC 涂于昆明种小鼠腹部致敏,6天后用PC 在肝脏攻击,测量血清谷丙转氨酶(sGPT)的活性并进行肝组织病理学检查结果发现,sGPT 活性明显上升,肝细胞坏死、门管区粒细胞和淋巴细胞浸润等。在PC 致敏前用环磷酰胺处理明显地增强了这种肝损伤。但给正常小鼠肝脏注射PC 则不能诱发肝损伤。这些结果表明,该肝损伤模型是因在肝脏局部发生的PC-DTH 所引起,其成立与DTH 反应的强度有关,可适用于肝疾病中肝损伤机理及治疗药的研究。
Picryl chloride (PC)—induced delayed type hypersensitivity (PC—DTH)was applied to miceto cause an experimental liver injury.The model was produced by PC challenge in the liver ofKunming strain mice that had been previously immunized with PC on the abdomen 6 days be-fore.The activity of glutamic pyruvic transaminase (GPT)in serum and histopathologicalchanges in liver were evaluated.As the result,distinct elevation of sGPT,necrosis of hepatocytesand infiltration of granuocytes and lymphocytes into the portal tract and sinusoid were observed.The pertreatment of cyclophosphamide before PC immunization markedly augmented the injury.However,the PC injection into normal mice liver did not induce an injury.These evidences sug-gest that the liver injury is a result of the PC—DHT occured in liver,related to the intensity ofDTH reaction,and this model is suitable for investigating the mechanism of liver injury as well asremedy for liver diseases.
