Ⅰ型嗜肺军团菌杂交瘤细胞株的建立和鉴定

摘要 用Ⅰ型嗜肺军团菌(Ⅰ型LDB)颗粒生抗原免疫的BALB/C小鼠脾细胞与小鼠骨髓癌SP2/0细胞在PEG怍用下融合,经二次克隆化后建立了一株分泌抗Ⅰ型LDB单克隆抗体的细胞株(No.19 McAb)。经ELISA、间接血凝试验、双向琼脂扩散试验、结合力试验,染色体检查及SDS-聚丙烯酰胺凝胶电泳的检测,证实此杂交瘤所分泌的McAb属小鼠IgG类,对Ⅰ型LDB是特异的。No.19 McAb将有助于Ⅰ型LDB的检测及其亚型的分析与流行病学调查. The spleen cells from BALB/C mice immunized with LDB I were fused withSP2/0 myeloma cells. A hyhridoma cell strain secreting McAb against LDB Iwas obtained after cloning twice. Its specificity was identified by enzyme-linked immunosorbent assay(ELISA), passive hemagglutination (PHA) and combining test. It was shown by double diffusion and SDS-PAGE tests that the monoclonalantibody belonged to IgG of mice. The monoclonal antibodies can be used as a reagent for detection of serogr-ouping of LDB I and investigation of epidemiological markers for LDB disease.
