摘要
应用篮式生物反器,以聚酯纤维为载体连续灌流培养乙肝重组CHO-C_(28)细胞,观察细胞生长和HBsAg的表达。结果灌流速度提高到每天3.5倍工作体积,收液中HBsAg的RPHA效价在维持期为1:256~1:1024。在细胞生长旺盛时,可提高转速使溶解氧(D.O.)控制在20%~30%。在收液中,由于细胞残渣少,既可避免灌流系统堵塞,又有利于提纯。
Recombinant CHO-C_(28) cells were continuously perfusing cultured in a Celligen-packed bed basket bioreactor using fibra-cel carrier, and cell growth and HBsAg ex-pression were observed. The results showed that the RPHA titre of HBsAg in harvested liquid was 1: 256~1: 1024 during maintaining period. Under this condition, perfusion rate could be improved to 3.5 working volumn/ day. When cells were growing exuberantly, D. O. could be controlled at 20%-30% by regulating stirring speed. Little cell residue was found in harvested liquid, so the blocking up of perfusion system could be avoided and purification was easy to carry out.
