牛副结核分枝杆菌基因文库的构建及特异性DNA片段的筛选

Establishment of genomic library of Mycobacterium paratuberculosis and screening of specific fragment of its DNA

以质粒pBluescript SK为载体,用Pst Ⅰ构建了牛副结核分枝杆菌C_2株的基因文库,并初步筛选出4个与牛结核分枝杆菌、禽结核分枝杆菌、草分枝杆菌不反应的重组子。快速提取重组质粒分析的结果表明,这4个牛副结核分枝杆菌的特异性DNA片段大小分别为5.1kb、2.2kb、1.7kb和1kb。

The genomic library of Myco. paratuberculosis strain C2 from cattle was established using plasmid pBluescript SK as a carrier by enzyme Pst I cutting. And four recons which didn't react with Myco. bovis, Myco.avium and Myco. phlei were preliminarily screened, the specific fragments of DNA from the four bacteria were 5.1, 2.2, 1.7 and 1 kb in size respectively by rapid extract recs-plasmid assay.
