摘要
利用边缘边虫DS-1株接种除脾易感牛,制备出补反诊断抗原。所制备的抗原具有良好的特异性和敏感性,并建立了总量为0.6ml的补反诊断方法。对试验条件下人工感染牛的补反检出率为100%,安全区无假阳性反应。人工感染牛第5天出现补反抗体,感染后20~50天血清抗体达到高峰,以后开始下降,7个月后补反抗体消失。所制备的补反抗原与双芽巴贝西虫、牛巴贝西虫、大巴贝西虫,环形泰勒虫、瑟氏泰勒虫、伊氏锥虫和日本血吸虫等几种血液寄生虫抗血清之间没有交叉反应。抗原保存期达一年以上。所制备的抗原达到了国外同类产品的水平。
The diagnostic antigens for complement fixation test (CFT) were prepared using the splenecto-mized susceptible cattle inoculated with the blood infected by A. marginale strain DS-1 and were used in CFT established by us. The results showed the detectable rate of artificially infected bovines was 100 percent, and the complement fixation reaction antibodies appeared in the cattle on the fifth day after inoculation, and the serum antibody titer reached peak value 20 to 50 days after infection but began falling after the stage of this time and disappeared in 7 months. There were no cross reactions between the CFT antigens and antisera of Babesia bigemina, B.bovis, B.major,Theileria Sergenti, Trypanosoma evansi or Schisiosoma japonicum, and when the sera of cattle from safe areas were examined in CFT using the antigen, the results all were negative for antibody to A,marginale, which showed a good specificity and sensitivity.
