金蒸汽激光治疗人肺腺癌裸小鼠移植瘤

摘要 应用肿瘤内照射和肿瘤表面照射两种方法在人肺腺癌裸小鼠移植瘤(lung adenocarcinoma xenograft,LAX-83)进行光动力学治疗(photodynamic therapy,PDT)的实验研究。结果表明PDT后LAX-83生长延缓。治疗后10天,肿瘤内照射组和肿瘤表面照射组之间的肿瘤体积和重量无显著差异。而在治疗组与对照组、单独激光组和单独光敏剂组之间存在显著差异。形态学观察结果表明肿瘤内照射组的病理改变比肿瘤表面照射组出现更早、更显著。后者肿瘤坏死深度在4~5mm左右,而前者则达10mm以上。光镜下发现肿瘤细胞胞浆、胞核浓缩,肿瘤细胞坏死。电镜检查提示线粒体受损最早。单独注射光敏剂和单独激光照射后均不产生肿瘤组织的显微结构和超微结构的改变。本实验证实PDT对人肺腺癌细胞的杀伤作用,为肿瘤内照射治疗的临床应用提供有益参考。 A study of gold vapor laser-photodynamic therapy (PDT) on human lung adenocarcinoma xenograft in nude mice (LAX-83) employing surface illumination and insertion illumination was described. The results showed that the growth of LAX-83 was hindered after PDT. The volume and weight of the tumors had no significant difference between two treated groups 10 days after PDT. There existed significant difference between treated groups and control groups (contol groug, single laser group and single photosensitizer group) (P<0.01—0.05). The results of morphological investigation demonstrated that the pathologic changes were earlier and more marked in insertion illumination. The depth of the tumor necrosis is about 4-5mm in surface illumination and over 10mm in insertion illumination. The concentration of cytoplasm and nuclei in tumor cells and cytonecrosis were observed under light microscopy. With electron microscopic examination, the earlier pathologic change Was mitochondria impairment. There were no pathologic changes in single photosensitizer and single laser groups. It can be concluded, that the efficacy of PDT in the treatment of human lung adenocarcinoma cells was confirmed by the experiment. This study will provide useful reference for the clinical employement of insertion illumination.
