摘要
目的 通过鼠痘病毒抗体的制备摸索鼠源性多抗的大量生产方法。方法 用完全福氏佐剂乳化的鼠痘抗原经腹腔注射免疫 10只昆明小鼠 ,共免疫 4次 ,其间腹腔注射一次液体石蜡。结果 试验中 7只小鼠产生腹水 ,平均每只产 10 6ml ,最高达 2 6ml,腹水总产量是血清总产量的 15倍。同一只小鼠腹水抗体效价在不同采取时间保持稳定 ,不同小鼠间腹水抗体效价差异较大 ,在 1∶32 0~ 1∶5 12 0之间。腹水通过 5 6℃ 30min灭活处理后 ,效价没有变化 ,灭活处理的腹水冻存于 -4 0℃的条件下 ,6个月内经反复冻融效价基本保持不变 ,未经灭活的腹水冻存后效价下降明显。结论 用腹腔免疫诱发腹水可得到产量较大 ,效价较高的特异性抗体。
Objective To develop a method of production of the polyclonal antibody and its preservation.Methods 10 KM mice were immunized intraperitoneally four times with an emulsion of Ectromelia virus in complete Freund's adjuvant,together with a single intraperitoneal injection of paraffin liquid.Results Up to 26 ml ascites could be obtained from a single mouse,the mean value of each mouse was 10.6 ml.The total value of ascites produced was 15 times greater than that of serum.The antibody titres in the ascites of different mice had some difference,from 1∶320 to 1∶5120.With inactivation at 56℃ for 30mins,the antibody titres had no change in 6 months.In contract,the antibody titre of ascites showed significant reduction without inactivation.Conclusion Specific antibody with large quantity and high titre can be obtained with this method.
