摘要
目的 对近交系小鼠体内受精的胚胎在三种不同处理温度 (2 0℃、2 5℃、2 8℃ )进行玻璃化冷冻保存。方法 不同温度下 ,胚胎在 10 %EG溶液中预处理 5min ,再投入EFS40玻璃化溶液中 1min ,平衡后冷冻。结果 与2 0℃和 2 8℃相比 ,三个品系小鼠的胚胎都在 2 5℃冷冻时获得最佳效果 ,解冻后培养观察发育率分别是 87.77%、90 0 0 %、87 18%。结论 该结果对近交系小鼠胚胎保存具有重要意义。
Objective Mouse morulae from \%in vivo\% were frozen by vitrification at 20℃,25℃ or 28℃ and developmental ability of thawed embryos was assessed \%in vitro.\% Methods At different temperatures, the morulae were pretreated in 10%EG for 5 min and equilibrated for 1 min in EFS40, then plunged into LN 2 for cryopreservation. Results\ The results of 25℃ were better than that of 20℃ and 28℃, the survival rate of BALB/cJ, TA2 ,T739 embryos was 87 77%, 90.00%, 87.18% respectively after thawed. Conclusion\ The result is very important for cryopreservation of inbred mice embryos.\;
