摘要 为解决使用细胞培养法进行甲型肝炎病毒灭活试验,经实验证明NJ-3株甲型肝炎病毒可在人肝癌细胞PLC/PRF/5株中快速复制增殖。用免疫荧光法检测,在维持培养5天时即可观察到细胞内典型的HAV特异性荧光颗粒,维持培养10天,TCID_(50)达10^(5.5)。该培养系统适用于消毒学试验。 In order to use cell culture method for carrying out experiment on inactivation of HAV, human primary hepatic carcinoma cell PLC/PRF/5, African green monkey cell BSC-1 and Vero cell were used for culturing NJ-3 and HM-175 strains of HAV. IFA method was used to examine replication of HAV in cells. The results indicated that HM-175 strain was weakly positive in 3 cell lines after 8 passages followed by maintenance culture for 28 days, while NJ-3 strain was obviously positive after maintenance culture for 28 days during first passage or after 8 passages followed by maintenance culture only for 5 days. When NJ-3 strain of HAV after 8 passages was cultured in PLC / PRF / 5 cells, the TCID_(50) measured with IFA method was 10^(-4.0) on 5th day and 10^(-5.7) on 14th day. This cell culture examination system requires shorter time and is more suitable for examination of medium and large samples than RIFC method, therefore its use in examination of HAV activity in disinfection experiment can be considered.
