RADIOIMMUNOASSAY OF ATRIAL NATRIURETIC FACTOR IN HUMAN PLASMA——A COMPARISON OF THREE METHODS

本文报道了用玻璃粉(Vycor glass)提取人血浆心钠素(ANF)的放射免疫分析方法(RIA)与血浆直接RIA法和Sep-Pak C_(18)柱提取的RIA法进行了比较。三种方法的ANF测定值有相关性,而以玻璃粉法简便易行。

A Comparatitve study of three RIA methods for plasma α-human atrial natriuretic factor(ANF) was conducted. ANF concentrations were measured in 85 normal subjects by direct RIA and RIA following pro-extraction of the peptide from plasma by either Vycor glass or Sep-Pak C18 cartridge. The value of direct assay (222.5±53. 4 pg/ml, (?)±s)was 6～10 times higher than those of pro-extraction by Vycor glass (35.5±10.8 pg/ml) and by Sep-Pak C18 cartridge (22.9±4.3 pg/ml). Studies on recovery and coefficient of variation (CV) of the three methods showed that recovery of the direct assay was the highest but with larger CV. The findings indicate that values of plasma ANF assayed by two pro-extraction methods are very close, however, RIA following pro-extraction with Vycor glass is similar and inexpensive.
