摘要
人工虫草菌丝经热水提取、乙醇沉淀得到多糖粗制品(CHWc),产率6.67%,再经DEAE-Toyopearl 650 M柱层析,即得多糖精品(CHWp),产率1.71%,采用凝胶过滤法测得其分子量为3.2×10~4。纸层析(PPC)和气相层析(GLC)分析显示CHWp含D-甘露糖、D-半乳糖和D-葡萄糖,其克分子比值为1.0:2.7:1.8。口服给予CHWc剂量在500 mg/kg时对正常小鼠无明显的降血糖作用,而腹腔给予CHWc,剂量在100 mg/kg和50 mg/kg时,对正常小鼠、四氧嘧啶糖尿病模型小鼠和链脲佐菌素糖尿病模型小鼠均有显著的降血糖作用,且呈现一定的量效关系。
The dried powder of cultural mycelium of Cordyceps sinensis was extracted with hot water and precipitated with ethanol to give the crude polysaccharide (CHWc), in 6.67% yield. CHWc was purified by column chromatography on DEAE-TOYOPEARL 650M to give the purified polysaccharide (CHWp), in 1.71% yield. CHWp was homogeneous on gel filtration and molecular weight was estimated to be Ca 3.2×10~4. CHWp consisted of D-mannose, D-galactose and D-glucose in the molar ratios of 1.0: 2.7: 1.8 by paper chromatography (PPC) of the hydrolyzate and gas-liquid chromatography (GLC) of the alditol acetate prepared from the hydrolyzate. CHWc when given by the route of ig at a dose of 500 mg/kg had no effect in normal mice, but when given by rotate of ip at a dose of 100 mg/kg and 50 mg/kg it had significant hypoglycemic activities in normal mice, alloxan diabetes model mice and streptozotocin diabetes model mice, and showed dose-effect relation to a certain extent.
