摘要
应用TT.APTT,TGT等方法观察了由猪皮分离的硫酸皮肤素(DS)对凝血的影响以及DS与肝素(UFH)的协同作用。结果显示,DS的抗凝作用比UFH弱;有较强的催化抑制凝血酶生成的作用;明显增强UFH的抗凝及抑制凝血酶生成作用;此外,DS引起血小板聚集作用明显弱于UFH.提示:DS有较弱的抗凝作用.并与UFH有明显的协同抗凝作用,它是一种有希望的抗凝药。
The effects of dermatan sulfate (DS) extracted from porcine skin on human blood coagulation and platelet function were observed by the thrombin clotting time (TT),the activated partial thromboplastin time (APTT), the thrombin generation test (TGT) and platelet aggregation. The results showed that the anticoagulant activity and inhibition of thrombin generation of DS was weaker than unfraction hep-arin (UFH). Antithrombin activity of DS is 1/60-fold as much as UFH, inhibition of thrombin generation of DS is 1/160 - fold as much as UFH. Moreover,DS augments obviously the anticoagulant effets and inhibiting thrombin generation of UFH. Antithrombin activity of UFH 0. 13 mg·L-1increase by 6-fold and anti-activated partial thromboplastin activity of that
increase by 24 -fold in the presence of DS 12 mg·L-1. The effect of inhibiting thrombin generation resulted from the mixture of DS and UFH, which dose is decreased by 50% (DS 20 mg·L-1, UFH 0. 13 mg·L-1)is equal to that of only one of both (either DS 40 mg·L-1or UFH 0. 25 mg·L-1). In addition,DS 50 mg·L-1can merely induce weaker platelet aggregation (MAR = 8. 7±4. 9%) but 25mg·L-1UFH can induce powerful platelet aggregation (MAR = 64. 2 ± 12. 4%) and there is no synergistic effect of platelet aggregation between DS and UFH. In conclusion,DS has a weaker anticoagulation and a synergistic effect with UFH apparently in TT, APTT and TGT. It appears to be one of the promising anti-coagulant agents.
