中药白术免疫活性成分多糖的研究

白术醇提后的药渣用水提醇析后,经硼酸型DE-52分离得到甘露聚糖AM-3。Sephadex G-200凝胶柱层析,玻璃纤维纸电泳证明 AM-3为单一多糖。AM-3的分子量为12 000。红外光谱示AM-3为β-构型糖苷键,且有甘露糖的特征峰。多糖水解物经薄层层析及气相层析,其组成成分为甘露糖。

The polysaccharide, named AM-3, wasisolated by ethanol precipitation of the hot water extract of Atractylodes macrocephalaKoidz, which has immunoenhancing actionwith subsequent purification through cellulosecolumn DE-52 and gelfiltration column onSephadex G-200. It was homogeneous asjudged by electrophoresis and gelfiltration. The average MW of AM-3 was 12000,which was estimated from the calibrationcurve, obtained by the gel chromatography ofstandard dextran fraction of known molecularweights on Sephadex G-200. AM-3 was ho-mosaccharide, UV and IR spectrometry indi cated that the molecule of AM-3 possesses abackbone of β-D mannan, and that the compo-nent sugar of AM-3 is D-mannose.
