摘要
荧光光谱法测定丁胺卡那霉素含量,操作简便灵敏,试剂价廉易得,可同时测定大量样品。线性范围0~10μg/ml(r=0.9997);批内,批间RSD分别为3.52%和2.94%。测定波长Ex415nm,Em475nm。适用于微量样品中丁胺卡那霉素含量测定。
A new method was used for the quantita-tive determination of amikacin. Amikacincan be determined spectrofluorimetrically by afluorescent derivative developed by condensa-tion of the primary amino groups with acety-lacetone and formaldehyde under acidic condi-tions (pH=2.6). A 2.0ml sample was placedinto a 10ml glass test tube, 2.0ml reagent wasadded, and the mixture was heated for 10 minat 100℃ in a water bath. 10 min later, emisionwas determined at 475nm using a wave-lengthof 415nm for excitation in a Shimadzu RF-540fluorimeter. Calibration curves proved a linearcorrelation between the concentration of thedrugs and the intensity of fluorescence. Themethod was caliberated in the range of 0~10μg/ml, and had a sensitivity of 0. 2μg/ml.
