摘要
应用未标记抗体桥联技术-碱性磷酸酶、单克隆抗碱性磷酸酶(APAAP),可以解决因血细胞胞含有内源性酶造成的非特异性染色的困难,可获满意结果。应用APAAP对34例献血员和16例患者的外周血和BALF的淋巴细胞标记CD_5、CD_4和CD_8,结果良好。
The use of unlabeled antibody bridging technique the alkaline phosphatase monoclonal anti-alkaline phosphatase (APAAP) makes it possible to solve the problem with nonspecific staining caused by endogenous enzyme of the cytoplasm of blood cellse. Satisfactory results can be obtained if factors influencing quality control e. g. optimal fixation, antibody dilution, incubation time and temperature have been tuitably adjusted. APAAP was used to demonstrate T-lymphocyte subsets of peripheral blood in 34 blooddonors and of BALF in 16 subjects as a study group. Labeling of antigens on T-lymphocytes with mouse antihuman monoclonal antibodies-CD_5 CD_4 and CD_8in our study showed an excellent results with reproducibility, It is demonstraed that APAAP provides a useful immunoenzymatic technique in the study of T-lymphocyte subsets of BALF in patients with ILD
