摘要
本文采用体外原代培养技术,详细观察和描述了大鼠胚胎下丘脑神经元在体外的形态发育过程,在国内首次建立了稳定的胚胎下丘脑神经元培养模型。结果表明:细胞培养4h,大部分细胞已贴壁;12h,部分细胞开始向外伸出小的突起;24h,部分细胞成为具有两个突起的神经元;3天时,神经元胞体增大,部分细胞为具有3个突起的神经元;1周后,神经细胞突起互相连接成网,细胞突起随培养时间延长而增多;第4周时,细胞开始减少,但仍可见较大的、具有多个突起的神经元。本文还对细胞培养过程中胚胎胎龄的选择、细胞分离的方法等问题进行了讨论。
The technology of primary culture was used to observe the developing process of hypothalamic neurons of 17~18 day embryos and the culture model of hypothalamic neurons was built. The results showed: after 4 hour culture the neurons adhered to the dish, after 12 hour culture the neurons began to extend small process, after 24 hour culture the neurons extended 2 processes, after 3 day culture the neurons become larger and possessed 3 processes, after 1 week culture the processes of the neurons interweaved and increased as time went on, after 4 week culture the numbers of the neurons began to reduce but the large neurons with processes were still seen. We also discussed the methods about the selection and the dissociation of the neurons.
