摘要
用连细胞斑片钳技术,在Wistar大鼠单个心室肌细胞上记录了T型、L型、B型钙通道的单通道活动。人参二醇组皂甙单体R_(bl)(250μg/ml)显著抑制3种类型钙通道的活动,使其开放概率减少与开放时间缩短,对通过通道的离子流幅度无明显影响。用电子自旋共振法测定了培养心肌细胞的自由基含量。R_(bl)(30μg/ml)显著抵消黄嘌呤(0.42mmol/L)-黄嘌呤氧化酶(5.3nmol/L)所致的自由基含量增多。
Single channel activities of T type, L type and B type Ca2+ channels were recorded on single ventricular myocardiocytes of Wistar rats, using cell-attached patch clamp technique. Panaxadiol saponin monomer Rbl 250μg / ml inhibited the acivities of the three types of Ca2+ channels significantly, decreased the open-state probability and shortened the open time, without apparent influence on the amplitude of ionic current flow through the Ca2+ channels. Electron spin resonance was used to measure the free radical content of the cultured cardiomyocytes. Rbl 30μg / ml significantly antagonized the increase of the free radical content induced by xanthine 0.42mmol / L-xanthine oxidase 5.3nmol / L.
