用高效液相色谱法测定儿童氨苄青霉素的药动学参数

摘要 本文报道用高效液相色谱法研究儿童时期氨苄青霉素的药物动力学。本法以β-羟乙基茶碱为内标物,C_(18)反相色谱柱,0.1mol/L 磷酸二氢钾/乙腈(92/8)作流动相,214nm 检测,方法学考查表明该方法灵敏度、精密度高,特异性强,可用于药动学研究和治疗药物监测。8名儿童静滴氨苄青霉素100mg/kg,药物动力学符合二室开放式模型。其消除半衰期t_(1/2)β1.07h。 High performance liquid chromatographic(HPLC)method for determination of ampicillin in capillary blood andpharmacokinetic investigation in children was reported.A reversed-phase C_(18) column and a 214 nm UV detector was used.Themobile phase consisted of 8% CH_3CN,92% 0.1mol/L KH_2PO_4.β-Hydroxyethyl-theophylline was used as an internalstandard.Protein was precipitated from serum sample with perchloric acid.Supernatant was injected onto the column.Theresults showed that the HPLC method was simple,sensitive,precision,and therefore be used for the study of clinicalpharmacokinetics and drug monitoring.Eight children were droped i.v.with a dose of 0.1g/kg of ampicillin,and thepharmacokinetics of ampiciUin was studied by the above HPLC method.The drug concentration-time curves were a two-compartment open model,The mean peak serum level was 199.58mg/L;The elimination half-life(T_(1/2)β)was 1.07 h andthe apparent volume of distributiom was 12.16L.
