花生未成熟胚培养与植株再生 被引量:9

摘要 采用MS、B_6、White、Norstog四种培养基及六种植物激素,配制成多种培养基,对授粉后20—25天的花生幼胚进行离体培养和再生研究。结果显示:MS、B_6为幼胚离体培养和发育较好的培养基。MS+BA1.0mg/L(单位下同)+NAA0.05+GA_20.02+CH600+PVP0.2%+蔗糖5%能有效促进幼胚发育成熟。小苗通过MS+BA2.0+PP_(333)0.2+CH600的壮苗培养后在1/2MS大量元素+MS微量元素及有机物+NAA2.0+BA0.1+CH400+蔗糖2%中培养,诱根率高达96.6%。小苗移栽14天内保持饱和湿度其成活率为80%。再生植株在自然条件下能开花、结实。 Four basic media,MS,B5 While,Norstog,six plant hormones with different combination and concerntration and various sugar and other nutrient components were used in this study for immature embryo culture and plant regeneration of groundnut.The results showed that MS and B5 were favourable media for immature embryo growh in viro MS+BA1.0(mg/L.the unit used below is the same)+NM0.05-GC0.02+CH600+Surcose 5% was favourable medium for immature embryo in vitro development 2MS+MS organic nutrient+NAA2.0+BA0.1+CH400+Surcose 2% could efficiently induce root system of the seedling.Condition of dark or dim light was of benefit to the embryo development.0.2% PVP reduced the cultured embryo brown ratio.95%-100% humidity in 11 days after transplantation could increase the number of survival plants.The seeds of regenerated plants were obtained.
