摘要
应用已知对皮肤有直接刺激作用的三种化学物溶液:70%二甲亚砜(DMSO)、20%苯甲酸(BA)和1%十二烷革磺酸钠(SDS)作用于体外液气界面原代培养的人的表皮细胞,探讨它们对细胞中乳酸脱氢酶(LDH)、谷丙转氨酶(ALT)和谷草转氨酶(AST)漏出的影响。结果表明,它们均使LDH从细胞内漏出,培养基中该酶活性显著升高。提示,培养基中LDH活性升高,可作为化学物质对皮肤表皮细胞有直接刺激作用、细胞膜受损的指标之一。
In this study,the primary generative epidermis cells of human skin cultured on the air—liquid interface were used.The epi
dermis cells were exposed to three chemical solutions:70%dimethyl sulfoxide(DMSO),20%benzoic acid(BA)and 1% sodium
dodecyl sulfate(SDS).The leaking out effects of lactate de hydrogenase(LDH),alanine aminotransferase(ALT)and aspar- ??tateaminotransferase(AST) from inside of human keratinocytes after exposure to three chemical solutions were observed.The
results indicated that these chemicals made LDH leakage from keratinocytes and the activity of LDH in the medium increased
significantly.It is suggested that the activity of LDH in the medium can be used as an index of direct irritation to keratinocytes
and cell membrane damage.
