THE 57TH AMINO ACID OF HLA-DQ BETA CHAIN AND THE IDDM SUSCEPTIBILITY IN SHANGHAI HANS

摘要 用聚合酶链反应(PCR)扩增了IDDM患者组和对照组DNADQEB_1位点第二外显子片段,并与SSO杂交。结果表明:分析DQβ链57位为非门冬氨酸(NA)/门冬氨酸(A)杂合子时DQ-α_(52)位为精氨酸或非精氨酸带来的易感性或抵抗性,可见白种人和黄种人IDDM关联格局是一致的;HLA-DQB链57位为A时与IDDM负关联,为NA时与IDDM正关联。黄种人中NA纯合子频率极低(0.16),可能是构成黄种人IDDM发病率远比白种人低的原因之一。 We amplified the HLA-DQB1 gene by polymerase chain reaction and hybridized with allele-specific oligonucleotide probes in 40 insulin-dependent diabetes mellitus (IDDM) patients and 42 normal subjects (the Han nationality in Shanghai). We found that extremely low frequency of non-Asp homozygotes in Chinese may contribute to the lower morbidity of Chinese IDDM than that of Caucasian IDDM. After combinative analyses of non-Asp/Asp in DQb57 and Arg in DQa52>we found a similar genetic pattern in Caucasian and Chinese IDDM and confirmed that Asp at position 57 of HLA-DQb chain centered resistance while non-Asp at this position confered susceptibility to Chinese IDDM.
