摘要
取灌喂克癌7851(160 mg/kg)20d的荷HepA肝癌小鼠的腹水癌细胞进行细胞生物学特性的研究,把经过一系列处理和荧光染色的癌细胞在FACS 420型流式细胞光度计上进行分析。结果表明:①克癌7851可使癌细胞的DNA含量明显下降(从1511.7±1.2降至133.5±4.4 AU或从157.0±1.7降至146.0±3.0 AU);②DAN指数(DI值)和增殖指数(PI值)亦明显下降(p<0.05和p<0.01);③可使癌细胞的G1期细胞数明显增加,从43.2±4.5%增加到78.3±5.5%,而S期和G 2M期的细胞数明显减少,而且DNA组方图的多倍体峰变得低平。提示克癌7851可以抑制癌细胞的DNA合成。
By flow cytometric analysis, cell biologcal properties of the cancer cells from BALB/c mice with ascites were studied. The results showed that: ①In AC 7851 treated group, the DNA content of the cancer cells decreased about 20 AU, i. e. from 151.7±1.2 AU to 133.5±4.4 AU or from 157.0±1.7 to 146.0±3.0; ②DNA index (DI) and proliferation index (PI) also decreased obviously (p<0. 05 and p<0. 01, respectively); ③The number of cancer cells of G1 phase increased from 43.2±4.5% to 78.3±5.5%, and that of the cells in S and G2M phases decreased. The polyploid peaks of the DNA histogram of the cancer cells in AC 7851 group became lower and flat, which suggested that AC 7851 could inhibite the DNA synthesis of the cancer cells.
