克癌7851对癌细胞的作用

摘要 经口连续给荷Ehrlich腹水癌的BALB/c小鼠克癌7851(AC 7851 160 mg/kg)15d,结果表明癌细胞死亡率明显增加,而腹水量明显减少,二者呈高度相关。AC 7851体外实验用扩散合细胞培养法,结果对空白对照组(不加任何药物)、阴性对照组(黄芪提取液0.6mg/ml)肝癌细胞无杀伤效应;而阳性对照组(阿霉素0.8μg/ml)明显地杀伤了癌细胞(死亡率为14.9±0.8%),AC 7851组(剂量为16μg/ml)也杀伤了大量的癌细胞(死亡率为6.9±1.3%)。扫描电镜下见癌细胞微绒毛大量脱落,许多细胞崩解破坏,破损的细胞粘连在一起形成团块。透射电镜观察到,胞膜、胞核、线粒体及异染色质等均有不同程度的损伤,乃至细胞崩解破坏,核内可见许多圆形的、电子密度高的致密颗粒,其杀伤效应有随AC 7851剂量增加而增加的趋势。在同样条件、同样剂量下,AC 7851对正常淋巴细胞损伤较轻。 In vivo, BALB/c mice bearing Ehrlich ascites tumor cells were administered orally with AC 7851,160 mg/kg for 15 days. Then, the volume of ascites and the survival rate of cancer cells were measured. The results showed that the volume of ascites in AC 7851 group reduced obviously, and number of dead cancer cells increased. Significant corelation between the volume of ascites and death rate of cancer cells was observed. In vitro,diffusion chamber was used for the cancer cell culture. Results showed that after 12 hours cultivation, the survival rate of cancer cells in control group did not decrease, meanwhile the death rate of cancer cells in negative control group (Astragolus Root) did not increased. Whereas, in positive control group (Adriamycin, 0.8μg/ml), the death rate of cancer cells increased significantly (the death rate. 14.9±0. 8%). In AC 7851 group (AC 7851, 16μg/ml), a large amount of cancer cells were killed. Under scanning electron microscope, the microvilli of the cancer cells disappeared, some cancer cells broke or burst, and many broken cancer cells glued to each other forming many clustters. Under transmission electron microscope, besides microvilli changes, injures of the membrane, nucleus, mitochondria and heterochromatin were observed. In addiion,many big or small,round electron density higher compact granules in nucleus were observed. While under the same condition and same dosage, the injury of normal lymphocytes was less. Therefore, it is suggested that direct killing effect is the important mechanism of anticancer effects of the AC 7851.
