麻竹离体快速繁殖技术的研究 被引量:41

摘要 当前我国发展竹笋生产遇到种苗供应紧缺的问题,而常规的育苗繁殖法劳动强度大、耗种多、运输不便、造林成本高。为了推广优良品种和培育大量的竹苗供群众造林用进行了竹类离体快速繁殖技术的研究。试验材料选用人工辅助授粉得来的麻竹种子和幼竹茎段。基本培养基用M_S,和不同浓度的生长调节物质组配,pH值近6.5,光照强度约1600 lx,每日辅助光照9~10h,温度控制在22~30℃之间。其试验结果是:芽增殖月平均2.3倍,试管苗生根率最高达93.9%,芽丛移栽成活率平均80%以上,试管外生根率66.0%以上。已有数以千计的芽丛移出试管外,苗长势良好。通过试验作者认为:1.采取两种培养基交替培养比用一种培养基更有利于维持无性系的生活力。2.IBA、NAA混合使用促进芽丛生根的效果比单独使用好。此外还观察到5月龄组培苗在试管里开花的现象,并对开花的原因进行了探讨。 Study on rapid propagation in vitro of Dendrocalamus latiflorus with seed obtained through artificial pollination and stem of young seedling, M_S as essential culture medium composed with different concentration growth-regulating substance, 6.5 of pH, about 1600 lx of light intensity, 9~10 h of daily supplementary illumination, 22~30°C of temperature showed that bud proliferation increased by 2.3 time a month, rootability of tube seedling reached by 93.9%, average survival of bud transplantation were over 80%, rootability of seedling out of tube reached over 66.0%. Flowering of 5-month tube seedling was observed and its causes were discussed.
