日本血吸虫24-26KD和90KD蛋白质“靶抗原”的抗原性 被引量:3

ANTIGENICITY OF 24-26KD AND 90KD PROTEIN "TARGET ANTIGEN" IN ADULT SCHISTOSOMA JAPONICUM WORMS

摘要 作者从日本血吸虫成虫抽提出两种国际公认在诱导保护性免疫力中起重要作用的24-26KD和90KD蛋白质“靶抗原”,用以免疫小鼠.结果表明上述两种“靶抗原”具有明显的抗原性.表现在(1)上述“靶抗原”免疫后可刺激宿主血清IgM升高,特别是IgG中的IgG1明显升高;(2)免疫鼠血清中的抗体能在成虫的表皮和实质层定位;(3)以ELISA和IFA检测免疫血清中的抗体水平时,均表现特异性抗体滴度明显增高(ELISA:90KD 1:5/20,24-26KD/:640;IFA:90KD1:80~1:2560,24-26KD1:40~1:160);(4)免疫血清中均存在着针对上述两种“靶抗原”的特异性抗体,免疫印渍试验结果进一步表明上述“靶抗原”免疫鼠血清中抗体均能特异地分别识别日本血吸虫抗原中分子量90KD和24-26KD的抗原决定簇。 Two protein 'target antigens' termed Sm97 and Sj26 which play important roles in inducing protective immunity can be extracted from S. japonicum worms. There are two bands in 24 and 26KD respectively for Sj26 and one main band in 90KD for Sm97 as identified by SDS-PAQE. Mice were immunized with these antigens and specific antibody responses in serum were assayed by ELISA,IFA, Western blot etc. The results show that: the antigens can stimulate mice to increase the level of serun IgM, especially IgG1 subset; the antibodies in serum of immunized mice can be localized in the antigenic determinants of tegument or body of the worms; specific antibodies against the antigens increased markedly after immunization as measured with ELISA or IFA(ELISA: 90KD serum 1: 5120, 24-26KD serum 1: 640; IFA: 90KD serum 1:80-1: 2560, 24-26KD serum 1: 40-1: 160); the antibody from mice immunized with 90KD can recognize antigenic determinants of 90, 148 and 40KD, the antibody from mice immunized with 24-26KD can only recognize 24-26KD antigenic determinant.
