赤豆叶肉原生质体愈伤组织再生植株 被引量:4

摘要 来源于赤豆叶肉细胞原生质体的愈伤组织,在含有3mg/12,4—D和0.2mg/l6—BA的D_(2a)或MS培养基上扩增培养约3个月,随后,转移到含有0.1mg/lIAA、O.2mg/1NAA和5mg/16-BA或无生长素而含有1mg/1 6-BA的MS固体培养基上分化长出了嫩绿的小芽,但在前一培养基中分化出的小芽不能进一步发育。当在后一培养基中分化出的芽长至3—5厘米时,将其从愈伤组织上切下,转移至无激素的MS减半培养基,或含1mg/1IAA的MS固体培养基中,经过一星期的培养均诱导出了根,从而形成了完整的植株。 <Abstract > After the calluses derived from mesophyll protoplasts of Phaseolus angularis had proliferated for about three months on D_(2a) or MS medium contain- ing 3mgl_(-1) 2,4-D and 0.2mgl_(-1) 6-BA, they were transferred onto various MS mediums for another three months. Shoot buds regenerated on MS medium sup- plemented with 0.1mgl_(-1) IAA, 0.2mgl_(-1) NAA and 5mgl_(-1) 6-BA or auxin-free MS medium supplemented with 1.0mgl_(-1) 6-BA. On the former medium the buds did not develop further. When shoots formed on 1after medium grew to about 3-5 cm in length, they were cut down from calluses and transferred onto hormone- free semi-MS medium or MS medium supplemented with 1mgl_(-1) IAA. Root for- mation was induced after 7 days and therefore complete plants were obtained.
