用聚丙烯酰胺薄层平板等电聚焦测定明胶等电点分布的研究 被引量:2

摘要 通过对聚丙烯酰胺薄层平板等电聚焦电泳的系统研究,建立了一套适合于分析明胶等电点分布的薄层平板等电聚焦电泳技术。实验结果表明,明胶等电聚焦要求相当低的凝胶浓度、较高的电功率和较长的电泳时间,这是由于明胶中含有较大的分子量级分及其松散的构象所致。在建立方法的基础上,测试了大量的照相明胶等电点分布,发现明胶分子等电点分布是由一系列不连续级分所组成,其中含量最大的主级分则对明胶的等电点及分布起主导作用。 In this paper the isoelectrofocusing on polyacrylamide thin slab-bed was studied sys- tematically and the method suited for analyzing photographic gelatin was established.It was resulted that lower polyaerylamide concentration,higher electric power and longer electrophoretic time should be used by reason of high molecular weight and extensive con- formation of fractions in gelatin and the resolution of electrofocusing pattern diagram was increased. A lot of photographic gelatin was research by the established method.It has been found that the isoelectric point distribution(IEPD)is uncontiuous.There was a group of peaks in the electrofocusing pattern diagram corresponding to a number of fractions with various isoelectric points.The main peak and its isoelectric point play a great deal to the IEPD of gelatin.
