高温和细胞外因素对某些类型恶性瘤细胞生长的影响

摘要 采用体外孵育,体内接种的方法,重点观察在细胞外因素作用下,41℃高温对几种瘤细胞生长的影响,从中找出了一些能增强高温效应的因素。通过~3H—TdR,~3H—UdR掺入DNA、RNA量测定,核仁内嗜银颗粒的检查及电镜观察,结果表明:41℃高温,细胞外高渗和低pH对瘤细胞生长有较好的协同抑制作用,这种抑制作用不受肿瘤细胞株的影响。 The combined effect of hyperthermia and extracellular factors on tumor cell growth was observed by using a method of incubation in vitro and inoculation in vivo.Several kinds of animal tumor cells were incubated in media of pH 6.8-7.4 and different osmolarities at 41℃ or 43℃ and were then inoject-ed into mice. It was shown that incubation of the tumor cells at 41℃ alone exerted little effect on their growth , while low pH or hypertonic media made the cells more sensitive to the inhibitory effect of hyperthermia. Thus, hyperthermia and simple medium of hypertonicity, or hyperthermia and low pH had more marked synergic inhibitory action on tumor cell growth. 3H-TdR aed 3H-UdR incorporation into DNA and RNA, nuclear argyrophilic granule examination and electronic microscopy of the tumor cells indicated that synergic action of hyperthermia and extracellular acidity or hypertonic medium of low pH was mainly due to inhibition of both synthesis of DNA and RNA, and inhibition of transcription of preribosomal RNA in tumor cells. These in turn affected tumor growth.
