慢性活动性肝炎细胞外基质的免疫电镜研究

Immunoelectron Microscopical Study of Extracellular Matrix in Chronic Active Hepatitis

本文利用Ⅲ、Ⅳ、Ⅵ型胶原及LM、FN抗体,对10例CAH肝穿刺组织标本用戊二醛多聚甲醛固定;NaBH_4恢复抗原性,厚片染色再包埋等作改良免疫电镜观察,结果较为满意。证实各型抗体在Disse腔均阳性,但强度各有差异。肝细胞之间只有FN阳性,其余阴性,意义未明。储脂细胞内质网Ⅲ型胶原、LM和FN阳性,支持该细胞是肝内产生间质的重要细胞。

Ten needle biopsies of chronic active hepatitis were studied by immunoelectron mi-croscopy using type Ⅲ,Ⅳ,Ⅵ collagen,laminin and fibronectin antibodies.We obtained good resultsby the immunoelectron microscopy(fixation in glutaraldehydeparaformaldehyde solution;restoringantigenicity by NaBH_4;cutting thick sections on HM881 device).All antibodies'reactions are positivein the space of Disse,but their positive degree is different.Between adjacent hepatocytes,there is onlyFN positive,all others negative;the significance is unknown.Type Ⅲ collagen,LM and FN are alsopositive in the plasmic reticula of Ito-cells.The findings support that Ito-cells are important cells forgenerating ECM in liver.
