胰岛素放射受体测定法和放射免疫测定法联合试验的初步评价

摘要 本文采用RRA法以检测生物活性胰岛素的变化。共检测IDDM及NIDDM 81例,后者并根据有无肥胖及病情轻重分成若干亚组,与RIA法作IRT的联合试验,显示NIDDM存在不同的胰岛素反应型及/或胰岛素相对不足。取部分病情较重的NIDDM患者作胰岛素红细胞受体各参数的测定,显示尚存在受体数目和结合力的降低。若RRA和RIA的结果显著不一,有助于发现活性异常的胰岛素类物质。以上结果证明,NIDDM是一组病因不一的异质性疾病。 A radioreceptor asssay (RRA) has been devised in our laboratory to test the changes of biologically active insulin. Eighty-one diabetes mellitus patients were examined, including IDDM and NIDDM; the latter has been divided into several subgroups according to the severity of the disease and the state of obesity. The combined RRA ond RIA has shown different reaction types towards insulin and/or relative deficiency of insulin among most of the NIDDM patients. The laboratory has also selected 5 severe NIDDM patients to determine the parameters of erythrocyte insulin receptor. The test demonstrated a decrease of the concentration of insulin receptor on RBC (sites/RBC) and/or post-receptor defects. This combined test is contributive to finding of the abnormal active insulin and insulin-like substances when there is significant difference between the results from RRA and RIA. On the whole, there is enough evidence to prove that NIDDM is a kind of heterogeneous disorders.
