同步化技术在白血病骨髓细胞染色体研究中的应用 被引量:1

摘要 采用改进同步化技术,即5-氟脱氧尿嘧啶核苷(Fdr)、尿嘧啶苷(Ur),加胸嘧啶核苷(Tdr)或5-溴脱氧尿嘧啶核苷(Brdu),对129例白血病和骨髓增生异常综合征(MDS)进行骨髓细胞同步化培养、染色体制备和显带,成功率达到95.4%,发现染色体畸变61例,包括78个异常克隆。本技术同一标本可用于G、R显带,与常规法比较,能产生更多有丝分裂细胞,染色体分散更好,显带较为细长清晰,且引起染色体损伤和丢失也更少。与其他同步化方法比较,免去洗涤细胞,更适合白血病的研究。 Chromosome study by using bone marrow culture with synchronization in 129 patients with leukemia and MDS from November 1984 to April 1987 is reported. Satisfactory cultures and bandings were found in 123 cases, a successful rate of 95.4%. Chromosomal anomalies were identified in 61 cases. Seventy-eight abnormal clones including numeric anomalies and structural anomalies of chromosomes were found. A method for synchronization of bone marrow cells with Fudr + Ur/Tdr or Brud was presented and compared with routine method. The technique was found more beneficial to bone marrow cultures. G and R-bandings could be obtained in the same preparation by this method. Generally it produced a higher mitotic yield and less damaged to chromosome. It also showed that the chromosome length was increased and the quality of metaphase spread was improved. We could recognize more abnormalities quantitatively and structural rearrangement accurately.
