摘要
自adw_2克隆中分离纯化乙型肝炎病毒全基因及“S”、“C”亚基因片段,制备探针后,对59例慢性乙肝患者的肝内病毒DNA进行核酸分子杂交研究。从冲发现30例肝内有游离复制型HBVDNA。29例肝内“S”、“C”亚基因片段探针杂交结果相同;但有1例仅有整合的“S”基因片段伴有复制型“S”片段,并未测到“C”片段。另对14例肝癌的癌组织也进行了HBV全基因及“S”、“C”片段核酸杂交研究。本文对“C”基因缺失与肝癌的关系进行了讨论。
Complete HBV genome and 'S' and 'C' subgenomic fragments were separately isolated from clone adw2 and labeled With ^(32)P as probes. Liver biopsies from 59 chronic hepatitis B patients were studied for the status of HBV genome and 'S' and 'C' subgenomic fragments. In 30 cases, free replicating form of HBV DNA was detected and among them 29 showed presence of 'S' and 'C' fragments. However, in one patient there was integration of 'S' fragment without detection of 'C' fragment. Fourteen hepatocellular carcinoma specimens were also studied for HBV genome and 'S' and 'C' fragments. The possibility of correlation between deleted 'C' fragment and development of liver cancer is discussed.
