摘要
本项试验对绿色木霉(Trichoderma viride)Sn-9106在几种不同培养条件下,经固态发酵生产的纤维素酶进行了研究。结果表明:液体纤维素酶最大活力出现在10%麸皮含量的固体培养基中,随着麸皮含量的增加,纤维素酶活逐渐减少,4种辅助酶中,半纤维素酶活力逐渐降低。淀粉酶和蛋白酶活力逐渐升高,而果胶酶活力变化不明显,固体酶产率及固体酶活性也呈下降趋势。在固体培养液中,不同氮源对纤维素酶生产的作用不同。在无任何外加氮源时,纤维素酶活性很低。电泳图谱上蛋白质的谱带也较有氮源时少;加入无机氮源促进了纤维素酶的合成,而有机小分子氮源则促进菌丝的生长繁殖。
This paper is about the quantity and quality of cellulase from Trichoderma virid Sn-9106 at different cultural conditions on solid medium fermentation. The maximum activity of liquid cellulase can be obtained from the solid culture medium which contains 10% wheat bran. In pace with the increase of wheat bran, the activity of cellulase and semicellulase are low, while the activity of diastase and proteinase are high, and the yield rate and activity of solid cellulase are low. The chane of pectase activity is not apparent. Various nitrogens in the solid culture medium have different effect on the cellulase production. Without any extra nitrogen in the culture medium, the activity of cellulase is low, and the kinds of protein are few. Inorganic nitrogen promotes the synthesis of cellulase, and low molecular organic nitrogen promotes the growth of hypha.
