摘要
本文用转化集落法研究水样中有机污染物对细胞的转化作用.水样取自黄浦江不同江段,水中有机污染物用大网状402和W2树脂吸附,然后用乙醚洗提、浓缩器浓缩备用。CHO细胞定量接种,37℃培养,有机污染物浓缩液加入培养液染毒16h后再换液培养10d,作集落计数和观察细胞的畸变,比较转化集落和对照集落的形态特征,实验发现:水中有机污染物会减少CHO细胞的集落数使转化集落数增加,同时呈剂量反应关系。结果再次表明:黄浦江水中有机污染物有潜在的诱变危险。
The method of transformed colony in stuidy of cell
transformation induced byorganic pollutants in water is presented.The water samples were
collected from differentareas of Huang─pu River.The organic pollutants in water were
absorbed by passingthrough column of macroreticular resin 402 and W2,then eluted with ether,
and enrichedin KD concentration vessel.The CHO cells were exposed to the extract for 16h
bcforc 1Odculture.The numbers of colonies were counted and the aberralions of cells were
observed.The charactristics of the transformed colonies were compared with the control.
Reductionof colonies,and increase of transformed colonies were notioed to be dose─
depandent.Theresults indicated the potential mutagenic risk of the organic pollutants in the
river water.
