摘要
人精子穿透去透明带地鼠卵试验中受精卵的制备一般采用压卵法,但该技术难度较大,不易掌握,丢失卵子的机会较多,而且在应用上受到局限。我们对这一过程进行了改进,采用甲醇、冰醋酸、水(5∶1∶2)混合固定液,固定、低渗同步法制备受精卵。经过多次试验表明,本方法稳定,重复性好。改进的方法不仅可用于男性学,生殖生物学等临床和研究领域,还可与分子生物学相结合、在分子水平上研究种间遗传物质的传递,减数分裂后基因表达的研究,非整倍体形成机制以及研究外源性因子如某些病毒和癌基因通过生殖细胞进入人类的机制等。因此,它在医学生物学,单倍体遗传学,以及人类遗传学等领域中具有不可估量的应用前景。
Abstract It was previously
reported ported that the preparation of fertilized ova from spermpenetration assay was not easy,In
the present study,a synchronous method of usingfixation with methanol ,acetic acid and distilled
water(5:1:2)to fix and expand fertilizedova was developed.The detections of more than 100 fertile
and infertile men showed thatthe stabiblity of the improved method is high,and the results were
alike to reportedmethods at home and abroad.This method uses not only in clinical study of
andrology,but in medical biology,its value of use will be inestimable.
